Sandman, hands down.
Swamp Thing and Preacher are also fantastic, and someday I'll start reading 100 Bullets, Fables, and Transmetropolitan, but Sandman redefined the art form.

Couldn't get into Sandman. Tried to read the second trade and stopped. Maybe it's just not my thing.

Sandman is going to take this. But I am voting for Fables. When you get down to it there are so many good ones to choose from. 100 Bullets. Preacher. It could almost be a tie between multiple books.

Preacher wins it with me....with Fables and 100 bullets following closely behind.

I went for Sand Man.
But love Fables, 100 Bullets, Preacher and pretty much everything else on the list.

Of the list, I've never read Swamp Thing, only read a handful of issues of Preacher, The Invisibles, Shade the Changing Man, Books of Magic, and Lucifer, and I've read at least a couple years' worth of everything else. Sandman is at the top of my list, followed by Transmet, 100 Bullets, Y, then Fables. I'm interested in reading or re-reading every book in the poll, though, which is a sign of just how good Vertigo is.

I gave it to Fables over Sandman, though it was really a toss up. I haven't yet read all of Swamp Thing, and I've read four volumes of Preacher. I'll finish Preacher and Swamp Thing, then maybe start another.
I read Y the Last Man but didn't see the big deal that everybody else seemed to. It was just okay.

Never really cared for Sandman. When it comes to Vertigo I have to give it to either Hellblazer or Preacher. Shade the Changing Man was also good, though it was sometimes hit or miss. Vertigo Doom Patrol was a lot of fun as well.

Sandman, Fables, Sandman, Fables, Sandman, Fables... Hmm... really difficult to choose one over the other.

Y The Last Man...because it's the only comic on that list that I've actually read.

Because I think Sandman is going to walk away with this, I'm going to go ahead and give Fables its due and give it a vote.

Should have left Sandman off the poll. It's going to win in a walk. I'd vote for it, too, but I will pretend it's not there and instead vote for Y: The Last Man.
Vertigo has had a lot of great series, though. Hellblazer, Preacher, 100 bullets, Fables, and more.

I've read at least some of everything on that list except Shade, the Changing Man.

Hellblazer - suffers from being an ongoing title with many creators, which means ups and downs in quality, and no definitive and final story arc to cement it in the minds of readers
Swamp Thing - Moore's run had an end, but Swamp Thing doesn't end there. The ongoing nature of the book again saps a bit from its overall ranking. Later runs were spotty.
Sandman - In many ways the definitive Vertigo book, at times being just about everything; magical realism, horror, fantasy, high fantasy, myth, and so on.
Transmetropolitan - A book I wanted to like, but didn't
Preacher - A core team of creators, a finite story that comes to an end, a concept not tied to other books. These are major pluses to an excellent book.
Books of Magic - At times good, but the quality varies. Hard to reach perfection when there are so many cooks.
Y the Last Man - Created with a vision from the start, a told a single and finite story, and left readers wanting more. A near perfect book.
Fables - Currently excellent, but in my opinion its ongoing nature will hurt it in the end.
Animal Man - Good when it's good, and a superb finish, but the art was never less than subpar and Morrison's writing lacks life, as always.
Shade, the Changing Man - DNR
Lucifer - Uneven quality. At times excellent, but too often unfocused.
Sandman Mystery Theater - Interesting enough, but can't play this game
The Invisibles - Overrated trash by a man who has read too much Robert Anton Wilson.
100 Bullets - An excellent crime book and a template on how to do a long run. Stable creative team, many smaller stories building into a single larger tale, and an ending. Great stuff.
